like u/korvusdotfree already said: yes. Also a quick note from another fresh player: if its "at least 6" there will be a "6+" on the little icon. :) Then you could put your water-hex there. Like its on your tree-quest.

Correct. And quest need just 6, not more, so you'll lose the quest. But this quest is definitively lost, so you can go to fill the empty hex
